Osteoarthritis(OA) is marked by inflammation, pain, and cartilage degradation, with elevated nitric oxide(NO) levels implicated in OA-related inflammation. We introduce a novel approach using a NO-scavenging hyaluronic acid conjugate(HA-NSc) with lubricating and anti-inflammatory properties for OA treatment. A derivative of o-phenylenediamine with NO-scavenging capability(NSc) was synthesized and chemically conjugated to HA. HA-NSc conjugate formed self-assembled nanoparticles(HA-NSc NPs) under aqueous conditions due to its amphiphilicity. HA-NSc NPs reduced NO levels and inflammatory cytokines under the activated murine macrophage model. In an OA animal model, HA-NSc NPs alleviated pain and reduced cartilage damage, demonstrating dual therapeutic action by combining HA’s lubricating property with NSc’s NO-scavenging ability. These results highlight HA-NSc NPs as promising agents for OA treatment, addressing pain, inflammation, and joint damage, suggesting their potential for OA therapy. |
